make this clear.. i too had this issue.. did u first connect a client computer to the domain before applyin the policies??? if yes, then the GPO policy takes into affect only after 16 hours(max)..if u want it to enforce immediately you do the changes in policies, then type the following command in the client computer command prompt..
    gpupdate /force

if not check the GPO whether it is linked and enforced..
    iPT 2G(S) - 8GB - JBn
    iPhone 3GS 16GB - JBn
    MAC OSx86 on P4 2.4Ghz
    HTC Touch - Elfin - P3452 - WM 6.5

did you apply the group policy objects to users or machines? also remember, no GPOs hit admin users... so even if the policies are being applied on the client machine, they wont matter if the user is a member of administrator, local or domain.
also, if you want to check if the policies are actually hitting the client, run rsop.msc (start -> run -> rsop.msc) this gives you the resultant set of policies... interface is identical to gpedit.msc... only the policies that are being applied to the machine and user are visible with their status (enabled or disabled)
do lemme know if this helped... otherwise will troubleshoot some more
